Sofronius berasal dari Damsyik. Ia mengembara kemana-mana, sampai akhirnya
menjadi rahib di Palestina. Sewaktu menjabat uskup Yerusalem, ia amat menonjol
sebagai pembela iman yang benar. Untuk itu ia menulis banyak buku teologi dan askese.
Ketika Yerusalem direbut oleh Khalif Omar (637), Sofronius memperlihatkan
tempat-tempat suci kepadanya dan berhasil mengambil hati Omar untuk berlaku
murah hati terhadap umat Kristen. Sofronius meninggal dunia pada tanggal 639.
